Chromatin remodeling: Activating ACL1 with a little help from 'friends'

ScienceDaily (Nov. 29, 2012) ? Chromatin remodeling -- the packaging and unpackaging of genomic DNA and its associated proteins -- regulates a host of fundamental cellular processes including gene transcription, DNA repair, programmed cell death as well as cell fate. In their latest study, scientists at the Stowers Institute for Medical Research are continuing to unravel the finicky details of how these architectural alterations are controlled.

Through a series of biochemical experiments, Stowers Investigators Ron Conaway, Ph.D., and Joan Conaway, Ph.D., and their team discovered that chromatin remodeling enzyme and suspected oncogene ALC1 (short for Amplified in Liver Cancer 1) is activated through an unusual mechanism: Its shape shifts in the presence of its activators. Their finding identifies a new instrument in cells' molecular repertoire of chromatin-remodeling tools and a potential cancer therapeutic target.

One of the main tasks of chromatin remodeling enzymes, Ron Conaway explains, is "to make DNA accessible so events like repair and gene transcription can occur." Postdoctoral research associate and first author Aaron Gottschalk, Ph.D., previously figured out that ALC1 required protein partners to activate its remodeling function. Publishing in an upcoming issue of the Journal of Biological Chemistry, he dissects the mechanism by which this occurs.

ALC1 and its ilk have a common protein domain, SNF2, that uses the energy of ATP hydrolysis to move nucleosomes -- the basic repeating units of chromatin -- around, in a process called nucleosome sliding.

"I was intrigued because ALC1 has a unique macrodomain not found on any other SNF2 family member," Gottschalk says. His interest was further piqued when he found that while most ATP-dependent chromatin remodelers function as large multi-protein complexes, ALC1 appeared to work by itself. At the same time, where most of its group-happy family members readily demonstrated nucleosome sliding activity in vitro, ALC1 was not only a lone ranger but also "completely dead on its own."

Gottschalk deduced that ALC1 may function independently, but it needs a boost from a couple of sidekicks: PARP1, an enzyme that responds to several kinds of DNA damage; and NAD+, the substrate by which PARP1 transfers chains of poly (ADP-ribose) onto itself and other target proteins, in a process called PARylation.

Only when PARP1 and NAD+ are on the scene does ALC1 spring into action, altering the accessibility of DNA by shifting nucleosomes around. Gottschalk's earlier findings were published in the Proceedings of the National Academy of Sciences in June 2009.

"We then extended this research," Conaway says, "and the upshot of our recent JBC paper is that ALC1 is likely activated through a series of physical interactions. ALC1's unique macrodomain can bind PAR, and protein-protein interactions also occur between ALC1 and PARP1." Gottschalk and coauthor Rushi Trivedi, a graduate student in the Biochemistry & Molecular Biology department at KU Medical Center, developed a novel footprinting assay that enabled this observation. Rather than merely activating ALC1 and moving on, the researchers found that the trio of PARP1, NAD+ and ALC1 hangs out in a stable complex.

"So PARylated PARP1 and NAD+ are allosteric effectors -- by binding to ALC1, they alter its state from dormant to active," Conaway says. "It's an interesting mechanism that's different from how most other chromatin remodelers work. It may also help explain other evidence that PARP1 has the ability to rearrange nucleosomes and reorganize chromatin; this could be one way by which PARP1 exerts its influence."

Apart from its role in modifying chromatin structure, not much is currently known about ALC1. It's regarded as a possible oncogene, being found in excess in hepatocellular carcinoma cells and because overexpression of ALC1 in mice induces spontaneous tumors.

PARP1, on the other hand, has attracted plenty of interest as a potential anticancer drug target, due to its importance in maintaining genomic integrity. For example, in breast cancer cells lacking BRCA1 or BRCA2 function, blocking PARP could effectively remove the cells' last line of defense against DNA-damaging chemotherapy agents. To date, no PARP inhibitor has made it past phase III clinical trials, but pharmaceutical companies continue to chip away at the challenges around optimizing this form of targeted cancer therapy.

"A better understanding of the in-depth biochemistry we're uncovering on ALC1 and PARP1 may, in the long term, ultimately lead to new or more refined therapeutic strategies," Gottschalk says.

Meanwhile, having observed that ALC1 comes to life upon interacting with friends, Gottschalk now wants to understand precisely how this activation happens. With a knockout ALC1 mouse model handy, plans are afoot to extend his studies to an in vivo characterization of a chromatin remodeler that boasts an illustrious family pedigree, yet stands apart in the crowd.

The short has some cool CG tricks and some deadly camera work. The action and dialog is a bit?dodgy?at times, but given the budget it?s?definitely?worth a watch.

We shot this piece on a ?shoestring budget?, using just one 5D-Mark II-camera (with Canon L-series lenses). Our very first mission was actually to help another company (Quixel) with some live-action shots/scenes to simply showcase their new texture-tool for 3d-models in games and movies, but instead we came up with this concept idea and created a full blown short film. Primarily, we wanted to do something visual and entertaining to simply present this concept and showcase Quixel?s tools.

We did almost everything our self (with some 3d/texture/sfx help) and we used close friends/family members to be ?actors?, including myself playing the main character. So I guess that?s why the acting isn?t top notch and why we didn?t use so much dialogue ;) BUT we also wanted to keep it very visual, we wanted to tell a story with pictures (and UI-text of course). That was a big challenge but I think we succeeded. In the end we?re happy with the end result considering the conditions and people really seems to like it! That?s awesome to see and very rewarding! / Jimmy & Eric?

Analysis: Confusing hard and soft power in emerging markets

LONDON (Reuters) - Deserting debt-laden, recession-racked North Atlantic and Japan for the fast-growing emerging market world may have been irresistible for some investors but many others still remain timid.

Why? It may be a case of "hard power" versus "soft power".

If investment decisions hinged solely on the former - measured by raw data such as economic output, population size and military spending - then convergence between rich and developing nation blocs is nigh on complete.

Adjusted for currencies' purchasing power, emerging economies on aggregate reached a "watershed" in 2012 and became as large as the rich developed world, estimates hedge fund manager Stephen Jen.

That train has long left the station, of course, and may well be only part way through its odyssey. According to Goldman Sachs, the giant economies of Brazil, Russia, India and China alone are expected to surge from a fifth of global output today to a third by 2020.

Using an index of "hard power" traced back to the early 1800s, with inputs such as iron and steel production, energy use, urban population and per capita military spending, Jen reckons dispersion of hard power between the top economies of the United States, Europe, Japan, China and Russia is at its lowest for 200 years.

But he reckons the world is still some way off equality of "soft power" - classed variously as the ability to influence ideas through culture, education, democracy or diplomacy but also capturing "intangibles" such as social justice, good corporate governance, transparency of law or open markets.

There's no direct gauge of this fuzzier "soft power". But if you take gauges such as Transparency International's "corruption perceptions index" as one possible component, then no major emerging economy comes in the top 20 apart from long-standing financial centers such as Singapore and Hong Kong.

Through investment eyes at least, the lags in this sort of "soft" development go some way to explaining why emerging financial markets remain far behind in terms of size, liquidity, depth, transparency and legal protections and why they have so badly underperformed of late despite all the "hard" metrics.

The lion's share of world private investable capital still originates in the western world. A recent study by analysts at TheCityUK showed that more than 70 percent of the almost $80 trillion of pension, insurer and mutual fund assets is still based in the United States, Europe and Japan.

And it's this relative gap on "soft power" that may still determine investment success or failure in the years ahead.

"This grating of the tectonic plates - with the real economies moving past each other but with the emerging world continuing to rely on developed countries' financial markets - will continue to create distortions and volatility in the global financial markets," Jen said.

These distortions have been evident for over a decade as booming China and other giants without adequate financial markets of their own banked their new-found and largely state-managed wealth in the west, exaggerated the credit bubble there and triggered sometimes violent and unpredictable capital flows.

SEISMIC REVERSALS

On a simple level, emerging underperformance of late may just be a function of an integrated world economy where no market is immune from shocks in its richest trading partners.

But even stock markets in the austerity-sapped recession economies of the euro zone periphery have as a group performed better than the BRIC equity indices for two years in a row now.

And while developed world equity at large is up 6 percent over the past two years, emerging markets are still down 10 percent.

There are many explanations, some related to the speed in which markets priced the long-term economic projections in the early part of the millennium with a 500 percent advance in BRIC markets between in the six years to 2007, for example.

This gets to the bigger worry of whether emerging markets are deep and liquid enough to absorb a wholesale shift of western capital without periodic seismic reversals that unnerve relatively conservative investors like pension funds.

In what Bank of England economist Andrew Haldane described last year as the "big fish, small pond" problem, rapid capital shifts quickly create overvaluation and he reckoned just a 1 percent shift in western pension funds to emerging markets was equivalent to some 10 percent of their entire capitalization.

So much so that even though investors in global equities would have increased money up to four fold over the past 20 years, equity investors who spotted China's explosive boom as early as 1993 are still in the red. Shorter-term, Brazil's real has lost 20 percent since March with no ostensible "crisis".

Emerging bond markets have done much better of late, up more than 15 percent in 2012 for example. But much of these are hard currency fixed contracts governed by New York or London law.

Citing demand for property in London or New York at the worst points of the recent global crisis, Citi Private Bank's Chief Investment Officer Richard Cookson reckons some simple rules apply that still make many wary of emerging markets.

"After decades of crises, you may ask what's with the emerging world? The answer is rule of law, property rights, corruption, failed institutions. How much of that has changed? Not much."

What's more, it's not at all obvious that the mega emerging economies like China even have the desire to move toward a center ground on financial soft power at least.

"China wants the prestige of a global power but not the responsibilities," said Michal Meidan, Asia analyst at political risk consultancy Eurasia. "They would be happy to be considered a global financial power but they are not ready for the responsibility of shoring up the global financial system."

Folks,

This is just a quick retrospective on the process of our wealth building. ?Briefly, we are on the road to?achieving?a net worth of $4,000,000 by the time we retire, if not sooner. ? One of the things we?ve?commuted?to doing is taking periodic snapshots of our progress by sitting down and calculating our assets and liabilities. ?In a nutshell, we?ve managed to increase our net worth pretty significantly over time, but we?ve also seen a great deal of fluctuation.

Here are a few snippets from years past if you are interested in seeing how our net worth had progressed. Just after we started blogging, and just before we were married, in April 2006 our?net worth was at $303k. We saw a quick 9% gain by the next month, in?May 2006 to $330k,?February 2007 at $308k?- due to student loans and wedding expenses,?March 2008 at $311k,?April 2007 at $329k,?June 2007 at $359k,?October 2007 at $383k,?May 2008 at $393k, then we finally broke?$400k in June 2008!

With the crash of the economy in 2008, our stocks also took a thrashing and by April 2009 we down to?$313k.?? However, the rebound was pretty quick and by December 2009 we were back up to?$389k. ?As you can see from our progress, we?ve both experienced steady gains in many ways, as well as taken the hits that life throws at you.[net+worth+graph.png] We weren?t tracking our finances as closely in 2010 and 2011 as James was in Afghanistan working for the Army. ?That said, our most recent update reflected an influx of family money as well as investment in several rental properties, bringing our total wealth to just over 850k.

Patient's own immune cells may blunt viral therapy for brain cancer

ScienceDaily (Nov. 25, 2012) ? Doctors now use cancer-killing viruses to treat some patients with lethal, fast-growing brain tumors. Clinical trials show that these therapeutic viruses are safe but less effective than expected.

A new study led by researchers at the Ohio State University Comprehensive Cancer Center -- Arthur G. James Cancer Hospital and Richard J. Solove Research Institute (OSUCCC -- James) shows that the reason for this is in part due to the patient's own immune system, which quickly works to eliminate the anticancer virus.

The findings, published in the journal Nature Medicine, show that the body responds to the anticancer virus as it does to an infection. Within hours, specialized immune cells called natural killer (NK) cells move in to eliminate the therapeutic virus in the brain.

The researchers discovered that the NK cells attack the viruses when they express specific molecules on their surface called NKp30 and NKp46. "These receptor molecules enable the NK cells to recognize and destroy the anticancer viruses before the viruses can destroy the tumor," says co-senior author Dr. Michael A. Caligiuri, director of Ohio State's Comprehensive Cancer Center and CEO of the James Cancer Hospital and Solove Research Institute, and a senior author of the study.

"When we blocked those receptors, the virus has more time to work, and mice with these brain tumors live longer. The next step is to block these molecules on NK cells in glioblastoma patients and see if we can improve their outcome," says Caligiuri, who is also the John L. Marakas Nationwide Insurance Enterprise Foundation Chair in Cancer Research. This study of cancer-cell-killing, or oncolytic, viruses is an example of the value of translational research, in which a problem observed during clinical trials is studied in the laboratory to devise a solution.

"In this case, clinical trials of oncolytic viruses proved safe for use in the brain, but we noticed substantial numbers of immune cells in brain tumors after treatment," says senior author and neurosurgeon Dr. E. Antonio Chiocca, who was professor and chair of neurological surgery while at Ohio State University.

"To understand this process, we went back to the laboratory and showed that NK cells rapidly infiltrate tumors in mice that have been treated with the therapeutic virus. These NK cells also signal other inflammatory cells to come in and destroy the cancer-killing virus in the tumor."

The study used an oncolytic herpes simplex virus, human glioblastoma tumor tissue and mouse models, one of which hosted both human glioblastoma cells and human NK cells. Key technical findings include:

  • Replication of the therapeutic virus in tumor cells in an animal model rapidly attracted subsets of NK cells to the tumor site;
  • NK cells in tumors activated other immune cells (i.e., macrophages and microglia) that have both antiviral and anticancer properties;
  • Depletion of NK cells improves the survival of tumor-bearing mice treated with the therapeutic virus;
  • NK cells that destroy virus-infected tumor cells express the NKp30 and NKp46 receptors molecules that recognize the virus.

"Once we identify the molecules on glioblastoma cells that these NK cell receptors bind with, we might be able to use them to identify patients who will be sensitive to this therapy," Caligiuri says.

For the average American household, that could result in an increase of $1,000 in taxes in 2013.

Tax holiday was designed to stimulate economy

FICA taxes, designated under the Federal Insurance Contributions Act (FICA), are taken on the first $110,100 of income. It includes a 12.4% tax that is paid into Social Security and another 2.9% that is paid into Medicare. While the self-employed are required to pay all of their FICA taxes, employees split the bill with their employer.

Ordinarily, workers pay 6.2% towards their Social Security contributions. But when the payroll tax holiday was introduced in 2010, that rate temporarily dropped to 4.2%.

The idea was to put more money into the hands of consumers in the hopes of sparking more spending, which in turn would contribute to economic growth. Philip Noftsinger, Business Unit President at CBIZ Payroll, says a reduction in the FICA tax rate was one of the easiest ways for Congress to send money to consumers during the recession.

Some members of Congress believe it has had little positive impact. Some also say it has been taking away funding from the Social Security fund at a time when it needs it the most. By some estimates, that figure could be as much as $150 billion per year.

?It hasn?t had the impact they thought it would and decreasing the input into Social Security at a time when output is rising, isn?t good,? says Noftsinger.

Mari Adam of Adam Financial Associates in Boca Raton, Fla., says no one wants to see their paychecks shrink but Congress, and many Americans, know it?s necessary. Unlike federal income, FICA taxes go directly to entitlement programs for retirement and medical care in your elder years. In many ways, these programs are a form of insurance, which makes the taxes almost a type of insurance premium.

?We?ll all complain but it?s the right thing to do. Social Security is already not on sound financial footing and it?s hard to argue that,? says Adam. ?We all need this.?

Impact on paychecks small but noticeable

News about the election and the overall impending fiscal cliff has overshadowed the fact that neither party has expressed an interest in renewing the tax break. Some economists and financial advisors believe there?s little chance it will be extended, so in all likelihood, the employee?s portion of the FICA tax will return to its normal 6.2%.

For an average worker making $45,000 per year, that?s $900 in a year, or $17.30 less they?ll bring home per week. It may not be the end of the world but at a point in time when prices are rising, incomes are already stagnant and investment portfolios aren?t doing that well ? every dollar counts.

?It?s not like it?s $100 per paycheck, but it will add up over the course of a year. It?s like a night out per month for a family of four. They?ll notice it,? says Noftsinger.

Because many Americans have become used to that extra money in their pockets, they?re likely going to see it as a tax increase if it hits in 2013. And if they?re not keeping up with the news and current affairs, they?ll feel a little shortchanged when they get their first paycheck in the New Year.

Tom Reahard is CEO of Paycheckcity.com, an online payroll solution, says many employers will likely notify their employees if it?s certain the tax break will not be renewed.

?My guess is this will run down to the last minute but most employers will probably notify their workers at some point,? says Reahard.

"I felt as though the environmental dimension of food needed a voice," Schenk recalls. Today, American Flatbread operates three popular Vermont locations, exports frozen pizzas nationwide, and is franchising its restaurant concept in other states.

But profit isn't Schenk's only priority: For more than two decades he has donated thousands of his flatbreads to the poor and sick. He's also held an average of eight benefit bakes each year to raise money for those in need, from the Boy Scouts in Vermont to earthquake victims in Haiti. He also serves on the boards of several nonprofit groups and promotes grass-roots causes by staging public events at his flagship restaurant.

Although Schenk's unorthodox, off-the-cuff style sometimes stirs controversy, friends and advocates say he is an uncommonly generous man who's willing to risk his reputation to stand by his convictions.

Schenk's career perfectly illustrates the "triple bottom line" business philosophy of social entrepreneurs who place equal emphasis on people, planet, and profits, says Enid Wonnacott, executive director of the Northeast Organic Farming Association of Vermont. "It really came from a very loving, big-hearted place," she says of his early work establishing American Flatbread. "He didn't think it was going to make him rich."

American Flatbread restaurants have a family-friendly atmosphere featuring a wood-fired oven and gingham-checked tablecloths ? and a mission that promotes tolerance, social justice, and environmental sustainability. Each week Schenk writes a dedication for his menus that explores issues of the day, and he often ponders the teachings of ancient philosophers.

The flatbread is occasionally served with political overtones. In December 2002, for example, it was baked with salt harvested from the Dead Sea ? half in Israel, half in Jordan. And in December 2009 it was topped with Afghan saffron as a gesture of support for Afghan poppy farmers trying to find alternatives to the opium trade.

Schenk's aim is to stretch customers' minds but not alienate them, he says. "When we do things that cross political lines and social barriers, we can open doors that lead to places we can't predict," he said recently in an interview at his log cabin overlooking Vermont's Green Mountains. "That is how we improve understanding of one another."

Schenk often leverages his prominence as a businessman to take public stands on issues ranging from nuclear waste to agricultural policy. He doesn't employ a fixed strategy, he says, but typically pens an opinion article in Vermont's largest newspaper, The Burlington Free Press, and later stages a benefit bake or public event to promote conversations and activism.

A prime example is an informal campaign he started in 1998 to bring local food into public schools. After writing an opinion article that stressed a lack of connection between local farms and school cafeterias, he held a benefit bake and donated the proceeds (about $800) to schools in Vermont's Mad River Valley. He's repeated the event every year since.

There are a few undeniable truths about matchups between the Green Bay Packers and New York Giants that fans and bettors alike need to know.

First, the Packers really don?t like it when sportsbooks make them an underdog. They have responded furiously with victory after victory lately. In 10 games in this role (they are 2.5-point dogs against the Giants in Week 12), the Packers are 9-1 ATS, according to Week 12 trends reports .

"Price Is Right" model wins $7.7 million in discrimination suit

LOS ANGELES (Reuters) - A former model on the U.S. daytime television game show "The Price is Right" was awarded $7.7 million in punitive damages by a Los Angeles court on Wednesday after suing the show for not letting her return to work after giving birth.

The amount was in addition to $775,000 in compensatory damages awarded to 41-year-old Brandi Cochran on Tuesday, after the jury in the case ruled that the producers of the show had acted with malice by not taking her back after her pregnancy.

The jury rejected Cochran's claim for mental hardship she said she had suffered.

"I hope my case will help other women in the same situation," Cochran told reporters outside the courtroom after the trial.

Producers for "The Price is Right" said they planned to appeal.

Cochran, who is married to soap-opera actor Dean Cochran, told the Los Angeles Superior Court the show's producers began treating her poorly after she told them in December 2008 that she was expecting twins.

The former Miss USA and Miss Teen USA told the court that once she was pregnant, producers made disparaging remarks about her eating habits and weight, and removed her from the show's website.

The show's comedian host Drew Carey backed the producers in testimony earlier this month.

Cochran worked as model on the show from 2002 until January 2009. Her son was stillborn in February 2009 and her daughter was born prematurely the following month. Cochran said she asked to return to the show, but was turned down in February 2010.

Cochran said she was encouraged to sue by actress Hunter Tylo's successful 1997 pregnancy discrimination suit against the producers of the TV show "Melrose Place," who fired Tylo after she announced she was expecting a child.

Beyond Structured Settlements: Calling Out Governor Cuomo

Doubling down on a recent investigative article he co-authored about Executive Life of New York (ELNY) ("The Complete ELNY Saga "), Bill Coffin, Group Editor in Chief for Summit Business Media?s LifeHealthPro.com, has published a follow-up op ed article ("Governor Cuomo, I'm Calling You Out") in which he urges New York Governor Andrew Cuomo to "make this right. Fix ELNY before its payees must endure their harsh payment cuts."

According to its Mission Statement: NYLB "acts for the Superintendent of Financial Services as the court appointed fiduciary and Receiver of impaired and insolvent insurance companies, to maximize assets and resolve liabilities, return rehabilitated companies to the marketplace or distribute the proceeds of the company in a timely manner to creditors."

Benjamin M. Lawsky is New York's Superintendent of Financial Services. Lawsky reports directly to Governor Cuomo and is currently a defendant in an ELNY class action lawsuit? filed November 8, 2012 by attorney Edward Stone and representatives of the Christensen & Jensen law firm.

Coffin's earlier co-authored ELNY article suggested the New York state government or the insurance industry could (and should) clean up the "ELNY debacle". In his most recent article, Coffin charges Governor Cuomo with personal responsibility.

"The simple truth", according to Coffin: "[Governor] Cuomo could fix ELNY if he wanted to. If he thought fixing ELNY would help his chances for a Presidential run in 2016, ELNY would already be fixed..... The insurance industry in the state could simply be ordered to pay that amount."

So why, Coffin asks, has Governor Cuomo not taken action? "Is it because [Governor] Cuomo cannot stomach the notion of making the rest of the industry in the state pay for the actions of an irresponsible few? ... [If] that is the case, then there is always the possibility of paying for ELNY with other general fund monies..... [A]t the very least, [Governor] Cuomo could ensure that any and all fines levied by the Department of Financial Services (Ben Lawsky) go first to ELNY before going anywhere else."

Coffin concludes his appeal to Governor Cuomo by observing: "There is a difference between doing the easy thing and doing the right thing. Show your constituents that you know the difference, or be called out as being just as much a part of this long-running failure as everybody else who has had a chance to fix ELNY and come up short."
